Abstract
Tenibs refer to tyrosine kinase inhibitors, which are small molecule drugs. Its role is to block one or more protein kinases, often used in the treatment of tumors, leukemia and other cellular abnormalities of the disease. Fostamatinib is a newly produced substitute drug and mainly used for treating the primary immune thrombocytopenia. It is prominent in the cure of primary immune thrombocytopenia. In addition, it is often used in the treatment of spleen tyrosine kinase (Syk)-related diseases. There is a wide range of market demand, so it is necessary to understand and explore the medicinal properties of fostamatinib. Among them, the synthesis process also plays a very important role. The different structure of fostamatinib has a profound impact on its efficacy. For example, compared with the prodrug salt, the prodrug salt hydrate has more ideal stability characteristics, so it is necessary to further study the synthesis method in order to improve its effect and productivity in industry. In this paper, three synthetic pathways of fostamatinib were analyzed. For example, the synthetic pathway that reacted only for 30 h had the highest efficiency and yielded 37.7%. In addition, this paper also emphasizes the application effect of fostamatinib, including pharmacodynamics and pharmacokinetics, safety and therapeutic potential. Fostamatinib is water-soluble, highly bioavailable, and metabolized in the liver. After two phases of clinical trials, it has been confirmed to be effective in dealing with primary immune thrombocytopenia and has less toxicity. As a new drug, fostamatinib has broad therapeutic potential, which is manifested in the treatment of diseases such as new crown respiratory failure.
