Digital holographic microscopy (DHM) offers a label-free and high-throughput imaging approach for biomedical applications. In this study, we present a protocol utilizing DHM to quantify the characteristic morphological changes of neutrophils during Neutrophil Extracellular Trap formation (NETosis). Our method provides a non-invasive and cost-effective solution, eliminating the need for time-consuming sample preparation required for traditional blood smears or fluorescence flow cytometry. By harnessing the unique capabilities of DHM, we aim to provide a tool for the understanding of various inflammatory diseases and to support clinical intervention decisions at the point-of-care.
